An evolving landscape: Innovations in variant classification to reduce uncertainty in clinical genetic testing

We will describe the current landscape of variants of uncertain significance (VUS) in hereditary genetic testing and the status of VUS resolution in a large-scale clinical environment. We will then evaluate different approaches for resolving VUS with an emphasis on the value of applying new methods for maximizing the use of existing and newly available data. We will explore various aspects of the Invitae Evidence ModelingTM Platform, with a focus on a more quantitative machine-learning enabled approach for objectively and systematically incorporating clinical evidence into variant classifications. We will conclude the presentation with case examples to illustrate the approaches discussed.
Flavia Facio, MS, CGC
Medical Affairs (Invitae)Flavia M. Facio, MS, CGC, is a graduate of the Sarah Lawrence College Genetic Counseling Program and is certified by the American Board of Genetic Counseling. Her work history includes serving as genetic counselor at Memorial Sloan-Kettering Cancer Center in New York City, and as Associate Investigator at the National Human Genome Research Institute in Bethesda, MD. Currently, Flavia is a Clinical Program Manager within the Medical Affairs Department of Invitae Corporation, where she works at the interface between new variant classification and interpretation technologies and their implications for clinical care. Flavia has been involved in various teaching engagements, and serves as Faculty/Supervisor in the Genetic Counseling Training Program of the Johns Hopkins University School of Public Health /National Human Genome Research Institute. Flavia is the immediate past Chair of NSGC’s Justice, Equity, Diversity and Inclusion Stewardship Committee and of the Jane Engelberg Memorial Fellowship Advisory Group.
Molly Stetler, MS, CGC
Clinical Science Liaison (Invitae)Molly Stetler, MS, CGC, is a graduate of the University of South Carolina Genetic Counseling Program and is certified by the American Board of Genetic Counseling. Prior to joining Invitae, she served as a genetic counselor in the child neurology department at Children's Health in Dallas, TX where she guided patient families through the genetic testing process and assisted clinicians in integrating genetic testing into their practices. For the past 5 years, Molly has been a Clinical Science Liaison for the rare disease genetic testing division at Invitae. She is passionate about engaging and educating medical providers across diverse clinical settings.
Sarah Hamilton, MS, CGC
Medical Affairs (Invitae)Sarah Hamilton, MS, CGC, is a graduate of the University of Wisconsin-Madison Genetic Counseling Program and is certified by the American Board of Genetic Counseling. Prior to joining Invitae, Sarah worked as a clinical oncology genetic counselor both at the New Zealand Familial Gastrointestinal Cancer Service in Christchurch, New Zealand and the Rocky Mountain Cancer Center in Boulder, Colorado. Sarah specialized in hereditary gastrointestinal cancer syndromes, with a particular interest in polyposis. At Invitae, Sarah has worked on the Clinical Consultation Services team supporting clients and patients and also assisted with several oncology-related product launches. She now works on the Medical Affairs team as a strategic commercial support manager for Invitae’s platform technology which encompasses variant classification and interpretation technologies and their implications for clinical care.
